POSITION RESPONSIBILITIES Mouse colony management (40%) Manage, maintain and breed various mouse strains that will be used for the research projects. Perform and track the breeding of mouse with spreadsheets and inventory of multiple mouse strains. Detailed genotyping of each mouse in the colony to ensure the production and maintenance of the required strains for the ongoing experiments in the laboratory. This task is to genotype and monitor homozygote or heterozygote mice that are genotyped through PCR genotyping with mice tissue obtaining by tail snips or ear punches of the mice. In-vivo/In-vitro preclinical experiments (40%) Perform mouse subcutaneous (SC/SQ), intraperitoneal (IP) injections, bleeding, sacrificing and harvesting organs after the mouse is euthanized. Will additionally be responsible for in-vitro experiments using mouse splenocytes and lymphocytes and serum, such as T-cell stimulation experiments and serum antibody level measurements. Additional experiments include: (1) Tissue culture experiments with cell lines, primary cultures, mouse splenocytes and lymphocytes, and peripheral blood mononuclear cells. (2) Peptide binding and inhibition experiments. (3) Molecular biology experiments using ELISA, PCR, qPCR, and Western Blotting. (4) Immunohistochemistry experiments. This task requires experience in basic lab techniques such as tissue culture, ELISA, PCR, Western blotting, and immunohistochemistry, as well as experience in conducting physiological experiments in mice. Laboratory management (20%) When necessary, the research technician will order supplies for the lab, and will help with maintenance of equipment. In addition, the research technician will prepare standard buffers and reagents used by the laboratory and organize, track and place orders and help with orders and animal protocols. The person will: * Perform a moderate number of simple or moderate technical procedures;* Require only infrequent guidance;* Frequently be responsible for ordering supplies;* Occasionally be responsible for requesting quotes and information for purchasing equipment for the laboratory;* Occasionally modify technical procedures;* Occasionally train or guide other lab members;* May occasionally operate, maintain and troubleshoot complex, high tech equipment;* May modify experimental protocols; Occasionally analyze and interpret data;* May occasionally make written contributions to papers and grant proposals;* An understanding of moderately complex scientific literature and moderate understanding of research goals and rationale is required. *May not be applicable. QUALIFICATIONS * Required to have at least a Bachelor's Degree in a related science or equivalent skills and related experience, and 2 to 4 1/2 years of related experience. Experience with mouse colony management, genotyping, and experiments is preferred.* Familiar with basic procedures involved in ELISA, PCR and tissue culture and handling chemicals and standard lab equipment preferred.* Good organizational and communication skills are essential.
